What if a griefer decides to start locking up things that don't belong to them?
I don't consider griefing potential a terribly valid criticism (anything can be griefer material if they're dedicated) but it's something to consider...
Obviously, yes, but not in adventure mode which is where I suspect this kind of device will be the most useful. You might set it up to where it can only be destroyed if the combination is set, but it would take a coder to tell if those kinds of ideas are piratical or not. It won't have much use in single player any way.
In adventure mode, the rooms themselves would be unbreakable. Just add an iron door. An effective lock with the key being redstone...Well, unless they've got a pick.

Some form of NPC modification where I can include walking, talking, living and breathing NPC's that I can customize skin wise, speech wise, and even function wise (do note I use MCPatcher HDFix due to the friendly HUD and the fact the owner and or creator of Forge is an extremely bias and rude person, plus I love the shading affect).
You can customize model, skin, size, AI behavior, stats (health, strength, range, etcetera), dialog, roles, jobs, faction systems (comes with the mod) and build entire questlines if you so choose using this mod: http://www.minecraftforum.net/topic/833003-151-custom-npcs-splanmp/
Want a shopkeeper? Done. Fast travel system? Done. Traveling minstrel? Giant dragon? Freakish spider-enderman (Ender model with Spider skin)? Have at it. There are a number of models, with each given a large handful of custom skins. You can mix-and-match skins and models (though this can have poor results if the skin just doesn't wrap 'round the model quite right.) And you can insert your own skins into the mod as well.
It's soon to be updated with customizable projectiles.
Oh, and it uses Forge. You can get over that though.
I can't build anything that would not make sense in real life. For example, I can't make any buildings out of wool because... That just doesn't happen in real life lol.

The door only breakable by whoever placed it? Okay...How about the wall? Ceiling? Floor? What if they blast it down with TNT or something?
Actually useful locks are rendered nigh-impossible due to the nature of the game, I'm afraid.
