I am looking to play with some of the 512 resolution texture packs, and feel I have a beefy enough computer to handle it. It has 16 gigs of RAM, and a video card that has 1GB on it as well. I don’t know all of the specs off hand, as I am at work.
My question is what would I need to do to run them? I currently run the 256’s with no problem, but when I went to load the 512’s they just hung up at the load screen. How long does it generally take to load up? Also, how much ram should I have devoted to it? I currently have 4G, which I set up in the profile manager, under the java console… should I bump that number up?
I read a few things about optifine… what is that? Do I need it for the most recent version of Mine Craft (1.7.4)? Any help would be appreciated.

1GB of video memory is going to be a problem if you're planning to use higher resolution textures, Minecraft is more reliant on that.

For every exponential "bump" of the resolution of the textures, it takes twice as much as before to load the textures, if that makes sense. 4G should be fine for all resolutions.

Optifine is a modification to the game. Read more on that here,
