I haven't installed the new snapshot yet, but I bet it would be quite a challenge. I can think of a couple problems that would arise where it would only be beatable by luck of the world generation.
here is what I can think of
1. impossible to go to Nether. No nether portals (although with trading the Nether isn't necessary since you can trade for eye of ender)
2. Stronghold could be unreachable. (no caves leading to it)
3. End portal could warp you too far from the land of the end.
4. no ores or materials. (although trading with npcs seems to take care of this one)
Is there anything in adventure mode to help get around that? Can you place ladders, or vines or anything? can you even place lava/water to make a cobble ladder or bridge, or nether portal?
I know adventure mode is for people to make maps based on it, but just like superflat there will be people who will try to survive a randomly generated world in adventure mode.

Actually I think it would be very possible. It would be very similar to the no crafting challenge Bluemagic has, except you would have to rely completely on looted or traded for items.
As you pointed out, if no caves from the surface actually connected to one of the three strongholds that could be an issue.
You can use an ender orb to teleport over to the main land mass in the end.

Lighting it could be difficult. You would need to either acquire flint to make a flint and steel or wood and use lava to set the wood on fire to light it. Using creeper explosions to attain materials would be an option.
I think the "Adventure Mode" is suppose to be for maps made by players and hopefully over the Summer Mojang will make some maps for us. Just my opinion.
Good points about the enderpearls! that solves the End problem (although I've also appeared below the End island, and had to dig up.
And yeah we will have to see what can and can't be done with lava/water.
As for the creeper mining, that might be the only way to dig to the end portal if there is no cave. do creepers do damage to blocks in Adventure mode? I would think that would potentially ruin adventure maps.
And yes I know it is not intended to be played in Adventure mode in vanilla and I am looking forward to adventure maps But this sounds like a fun challenge too.

Can someone with the snapshot confirm that in adventure mode creepers explosions still damage blocks?
Also can you still use a bucket to pickup water and lava?

Even if that works. it will still be difficult to make a Nether Portal without being able to place blocks for a mold but not impossible!

I have a few suggestions for those who are willing to try this challenge:
players can explore worlds and check if there are ways of getting into the stronghold and reaching the end from the landing point.
If these 2 are ok, one can finish the game in adventure mode.
The next step is exchange the seed you already checked and get one checked by another player. This way you know in that seed you can reach the places you need without knowing exactly the locations or any other surprise.

As of the last snapshot, you don`t need to go to the Nether to get the ender eyes.
Therefore, you can buy them from the Villagers and place them to open the portal.
edit: actually, I think this is since 12w21a. Dunno.
It's actually really easy to make a nether portal frame using a bucket. Here is how. First dig a 1 deep pit that is 4X2. Make a wall along one side of the pit that is 4X4. Place water source blocks on top of the wall so that the whole wall has water flowing down it and into the pit. Then using the bucket, place lava source blocks against the wall starting from the bottom in the pit and working up the wall. The lava placed in flowing water instantly turns into an obsidian block. I make the pit 2 wide so that water doesn't want to flow all over, but just into the pit.

And rodobon remember in Adventure mode we can't dig but if pails work we could build cobblestone to simulate what you are talking about.

I`ll check some seeds too.
Other option is to force the entrance to the stronghold, and making a path for the battle in the end, then upload the map for someone to try it on adventure mode. This is an option to make some adjustments for other players.
It would be rather hard to start trading, because you could only really pick up mob drops to trade with. You'd also better hope that village you find has a Library, as there's no other way to have a Crafting table (that I can see).

With the new trading, it is possible. You can live in a village and buy some swords, kill endermen, create a nether portal using buckets, craft ender eyes with the village's workbench (library), find a stronghold and... Hm, find creepers to explode and dig down to the stronghold :3

I've always said that I'd like to see half-finished Nether portals randomly generated here and there, though.
