I, unfortunately, have a lot of these moments of frustration. Whether it's losing diamonds or losing a castle to a griefer, what single moment of Minecraft makes you want to grind someone's bones?
Whilst in the end, getting spawned on that stupid floating platform , only followed by the enderdragon swooping in and killing you, then falling into the deep dark void, and your items to never be seen again
A creeper once spawned on top of a house I had worked on for hours. I promptly ragequit and deleted the world.

Wait... are you offering to tell me why dropping zombies 23 blocks takes them down to 1 1/2 hearts, but 24 kills them? I want these stupid zombies to fall enough to let me punch them once to death!!!!!!
Was swimming in diamond armor, went afk and drowned then later I walked into lava in my stripmine attempting to get new materials and then I threw my brand new silk touch book into my lava trash can
Lol! So many of those moments to choose from. Most recent ones seem to be related to buggy mob spawning since the 1.7x updates.
For instance, I have an XP farm I built around a zombie spawner I found in my mine. If I save my game while there are zombies on the killing platform, when I load the game again, there will be a bunch of zombies attacking me. It appears the mobs spawn before the walls, and when there are a lot in one place, a bunch escape before the walls form.
I also get the occasional mob spawn in conditions they should NOT spawn in. Like in broad daylight, and over water. I've been playing Minecraft for a few months now, and my main base is on an island that is (or was) totally secure. It is far enough from shore that no mobs can track me, so they don't swim over, and it's very well lit, so no mobs spawn there. At least before the 1.7x updates...
A few weeks ago I was working on something on my island base when I saw a zombie appear on the water. As I was watching it move towards me something hit me from behind and I turned to see a spider. This happened in the middle of the day. Bright light, no shadows.
A few days ago, in broad daylight, I was working on something near my house, when [SSSSSS - BOOM!]. Again, middle of the day! I turned around to find the corner of my house gone, and in its place was a large crater full of DIAMONDS! The corner that the creeper blew up happened to have my chest full of diamonds there.
This is very frustrating because before the 1.7x updates my island was safe and secure. Now I have to worry about random, buggy mod spawns.
As far as "normal" game play goes... I visited a friend's world to help her out. She needed stuff from the Nether, but was afraid to go there. She's a builder, not a fighter. So I told her I'd go and get her stuff (Quartz, Blaze Rods, Ghast Tears, etc.).
Spent a bunch of time and energy mining diamonds and creating enchanted armor and weapons, then I was ready to rock the Nether!
Just a few minutes after arrival I came across 2 Ghasts that seemed to be trapped in a narrow space. Score! Sitting ducks for the taking! I dug a trench around them to get into a better firing position, readied my one shot kill Unobtainium Bow, popped up and shot the first one, then ducked back down into my trench as the fireballs whizzed by. One of those fireballs hit the Netherrack above my head and opened up the bottom of a lava lake, which promptly relocated itself into my trench (and much of the surrounding area).
That in itself would be pretty frustrating, but what made it worse was I had a potion of fire resistance that my friend had gotten off a witch she had killed, but I forgot I had it, and never drank it. D'oh!! She was not too happy about me losing all that diamond stuff and her one and only Fire Resistance potion.
However, I redeemed myself by going back to the Nether with lesser armor and weapons, and returning safely with a crap-load of Nether Quartz, a bunch of Blaze Rods, Nether Wart and Soul Sand, some Magma Creams (doesn't that sound like a good name for a Hemorrhoid treatment), and even some Ghast Tears. Oh, and I scored her a couple of Whither Skulls too!

When i'm on a server for like 2 years with other people ,Having fun, helping out people, and building a awesome city.
BUT one day the admin went crazy and griefed and destroyed everything we worked on then he shut down the server

The most painful, annoying, agonizing thing?
Well, that's a really hard question. I could make a small list of the most annoying things, but there was never 1. Not trying to boast here, (there have been a bunch of alpha players so meh) but I've been playing since early beta, but even then, I still have gone through a lot of agonizing things. However, my pain is actually limited for my time. I play with keepInventory on :3 (Yeah, I suck)
1. When my favorite server updated to beta 1.8. It became a pure survival server, but it used to be a half-survival half-creative server, but creative mode had came out, so that forced the server into pure survival.
2. Losing my classic world. It was nothing much, but it held such memories...
3. Rubbish administrators everywhere. My damned god, some of them think they own a server due to their rank....

Forgetting to bring a few potions of Fire Resistance when venturing in to the Nether, I either: A] Fall in to a lake, or pool, of lava while mining for resources, or: B] Get blown up and set on fire by a group of ghasts.
I had just existed a cave branch, and had entered the central ravine the other caves nearby were connected too. I had been mining for diamonds for my town to fend off a siege that would take place in a few days (On the server the owner attacks towns with hordes of mobs, with awesome rewards for surviving). The pickaxe I was using was lent to me by a friend, second in command of the town, our military commander. It was a [LEGENDARY] Unbreaking X Fortune X Efficency X Diamond Pickaxe that we had discovered in a dungeon the owned built under our house (it was named Yuma's Gaea Eradicator, in honor of my commander, who's named Yuma in the roleplay).
There was a thunderstorm, and I was heading towards the ladders leading out of the ravine. I must have had nearly three stacks of diamonds. A creeper fell off the ravine, and I leaped out of the way. Just before it landed, lightning struck, and it was supercharged. The blast killed me, and by the time I had arrived back at the ravine, the items had gone. I had gone from enough diamonds to last a month to none.
The city was razed. The remnants of our civillization fortified a village, with only two residents remaining (the librarian and the priest). The only bad thing is, a longtime enemy and griefer may know our location.
My laptop crashed with all my minecraft worlds and now i lost them,
i was working for a long time on an adventure map and it was almost done, also my survival world in which i had everything I could ever need
3 days ago found my first abandoned mine and , lost most of all my resources plus the ones i dug out there to mobs!! finally got home without being dead! but the amount of stuff i lost was maddening!!
On one of my worlds, I really needed an xp farm so that I could enchant my armor and tools. I spent a lot of time trying to find a dungeon, and ended up using X-ray to find one. When I got down there, I discovered that it was a skeleton dungeon (very valuable for arrows and bones). A creeper found its way over to the spawner, got shot by a skeleton, and blew up the spawner. I now have no reliable way of getting xp or arrows.
Most frustrating? Ha ha well have I got a story for you... but anyway it has to be when I make a house get settled get some tools! everything is fine then out of nowhere a creeper blows me and my base up and i lose it.. forever
