There is a small lake between 1 & 2 blocks deep next to where I am and there was plenty of squids spawning in it so I took advantage of it by killing them every time I went past. After acquiring several stacks of ink sac's I stopped to focus on other things.
Now I notice they are no longer spawning, would this be due to the cow pen I have constructed nearby or possibly the sugar cane farm with numerous water filled blocks?
I know that once you reach a extremly high number other peaceful aminals will not spanw if the chunk that in you case the cows are in is loaded, but that rarely happens. Do you have a other water area near you?
There can only be so many entities in the chunks you have loaded at a time. Basically, you filled up some empty entity spaces by having a cow pen. So there's less spots for squids.
I completely destroyed the sugar cane farm and filled in the water blocks. I killed all the cattle leaving just 3 each of chickens, pigs, cows and sheep, then moved their pens a fair distance away (there was far more roaming cattle in the area when I found the location). I have slaughtered pretty much all roaming cattle but still no squids, ANYWHERE.
None in the ocean, none in the swamp and none in the lakes, most of which I have now filled up.
Would an infinite village cause them to stop spawning? There is only a few villagers in it. I move them via water stream about 70 blocks away for trading.
Well, I have travelled at least 2000 blocks in every direction. Not a single squid was found. I'm beginning to think that they have completely despawned from my world.
I might try reinstalling but obviously keeping the save. Librarian trading is pretty pointless as it stands.

Just asking, but what villagers do you get in your village? You could try breeding them to get new villagers that could potentially help you.

The usual kind, but to able to trade with librarians and get the rare book trades it makes things quite tedious when their next trade is for written books and I have no ink sacs.
Also written books, whilst being a bit of a pain are worth 1 emerald each
On my quest to reach a Stronghold I was taken almost back to my original spawn point (between 6000-8000 blocks away).
A couple of days ago my daughter joined my game on LAN but quit because the spawn was miles away from me, thinking about it now this could coincide with the missing squids.
Anyway, after traveling all that distance, still without seeing squids, but realising how close I was to my original spawn, I told her join the game and I would find her.
Upon finding her then quitting, the next time I started the game... SQUIDS!!!

On Topic: I don't know much about spawning mechanics. You could move the lake somewhere empty, then expand it.
