I'm posting this mainly because the players on my server are losing interest in the server. You know the drill, "This server is boring, I already have infinite diamonds! What do I do now?" Of course, this happens in single player too, so I'm also posting this for myself. How can you put the fun back in Minecraft after you've already mastered everything?
I say, throw away the diamonds (or lock them somewhere where you'll probably forget them) and start anew with the same house, Or just plain leave everything at home and move to a new location! You'll probably get lost along the way and never find your way back! =3
Level 50+ is easy now because I have Etho's Blaze Farm. The End is impossible. Mods usually don't add too much, just lag the game more, or get boring after the first day.

Make a city within a 400 block area.
Minecraft is only as fun as you make it. Luckily, there's infinitely many things to do. Therefore, you'll never stop having fun with this game unless you really try to dig yourself a hole in the cosmic fabric of the universe and hide in it. And if you really cannot find anything to do, stop playing for a while. You'll think of something. Recently, I started on two more stories to my house. I'm thinking of building yet another house some chunks away, maybe underwater if I don't want to stay absolutely legit.

You have to destroy about twenty or so dragon-healing blocks on top of nearly-unbreakable pillars while nearly every block you place gets destroyed by a "dragon" that gives huge knockback and lots of damage all the while dodging placing your mouse on any of the Endermen, who will teleport to you and hopefully not kill you as soon as you do.
I take that back though, there are good mods. Just they are far and few between.

It's inevitable, everyone will loose interest in anything over time, if you want the thrill back then have some sort of event or goal for your players..like for example, a maze that's 500x500 with 3 floors no lighting with mobs inside, or maybe a contest or something, implement faction wars and make 3 seperate worlds (2 for the faction worlds and 1 for the battlegrounds or the neutral world) hire yourself a group of PKers (Player Killers) and let the players have the thrill of being hunter down by these crazy men, Build an epic stronghold on the surface with a throne room at the farthest part from the entrance and whoever can get there without dying can have the castle for himself (mob spawners hidden inside the walls and no lighting...also no placing blocks or destroying blocks)...if none of those can spark some motivation then I don't know what
Even with open-ended games, this is bound to happen eventually. If it isn't too destructive to the server, you might want to consider resetting it and starting over from square one, much like creating a new world in SSP. If you can't do this, I'd suggest doing something I've done sometimes on a few servers - throw everything you own into a lava pit and enjoy the challenge! XD
For single player, try some challenges, play a few custom maps, make a redstone/noteblock thingo, make a gigantic castle legit, play creative for a while, make your own custom map, install some mods, only eat zombie meat, etc etc. Thats the great thing about minecraft, the possibilities are endless. If you really can't think of anything to do, stop playing for a month or 2, play another game or something. Just try, I'm sure you'll think of something to do.

I use construction to break my boredom. It creates an immidiate necessity for resources (giving me reason to mine for more) and it's something you can do forever.
I think your members should go start a new town several days travel away from your current location, and build a 4 metre wide cobblestone road leading the way, with rest houses for night.
Enchant everything with level 50+?
Install mods and do crazy stuff?
