My hotbar and inventory for that matter is always extremely organized otherwise it drives me crazy. I suppose there's some sort of compulsion related to that but anyways mine is as follows:
Slot 1: Axe
Slot 2: Mining Pick
Slot 3: Shovel
Slot 4: Empty
Slot 5: Empty
Slot 6: Empty
Slot 7: Half the time Empty and the other half of the time there is a bed here.
Slot 8: Compass
Slot 9: Torches
My empty slots change depending on what I'm doing in the game, but slots 1-3, 8, 9 generally don't change ever.
I have two
When Mining:
1:Sword
2:Bow
3:Stone Pickaxe (I like to preserve precious material)
4:Iron/Diamond Pickaxe
5:Dirt
6:Porkchop
7:Free
8:Torch
9:Clock
Always like this:
1. Torches. (Makes sense for me. While walking around, exploring, this is the item you will use the most. Also, it is what I have selected when I'm not using any item)
2. and 3. Cobblestone and Dirt. (The blocks you place the most when mining or exploring. Also the most expendable blocks. Of course, when I'm building something, I replace these with the blocks I use the most for the respective building)
4. to 6. Pickaxe, Shovel, Axe. (Always in this order, makes it easy to select.
7. to 9. Free slots, filled up according to situation: For exploring or hunting, weapons and a compass, usually. For mining, workbench, furnace and a bucket of water. For building, blocks I need to build.

7. Sandstone (for laying circuitry on, and visibility)
8. (Bucket o') Water/Lava
9. Source of Light
