Adventure mode sounds great, and I know that a lot of people have looked at it and said "RPG!" Well, I like that idea but I'm not concerned with it being that complex, although the option would be nice.
Red stone circuits are great and the existing blocks are great; but it needs more, and the command block doesn't cut it.
For starters, we need more visual customization. Regardless of my personal, oft-mentioned "slime" idea, we need colored water. We need dye-able blocks. Essentially, objects need to be converted to greyscale with applicable colors, like how bione-specific coloring and sheep and wool work.
We need an "empty map" mode for free building and an biome controller for the same mode.
We need full control of monster spawns; initial, respawn rate, special actions or types, etc.
And, to tie it all together, we need a full scripting engine with many, many, many events and triggers, including custom quest items (if you don't know what I mean, go play with Staredit or RPGMAKER2k or 2k5.)
As it is, Adventure Mode is great for getting from point A to B with whatever the game engine randomly throws at you; with this, it would TRULY enable adventures to be built.
The ideas here aren't bad, up to the scripting engine thingies.
A modding API should be enough for fully customizing adventure maps. The recent superflat additions would be enough for your requested "empty map" mode.

Texture packs are a godsend. The greyscale idea, while sounding hunky-dory is actually murder on most common machines. Keep in mind, looking at the snooper information available from Mojang, the vast majority of users don't have computers with the latest graphics capabilities. The dyable blocks you're suggesting will kill an already filling terrain.png we just don't have the space available for it. They cannot work like leather armor and would need to work like the existing wool/sheep which means a whole new block for each color variant. In short, no support for this portion.

Spawner Customisation: I support this idea, I'd love to see a redstone activated spawner that isn't a dispenser with prefabricated eggs in it. I'd love to see access to a GUI to adjust spawnrate, HP, armor, weapon, drops and a variety of other options.

No. No. A thousand times no. Welcome to Creative Mode and Advanced Redstone Engineering. Please, go play some of the Adventure Maps created since 1.4 was released. A LOT of what you're suggesting here can be achieved with redstone, naming, lore, enchanting, and a number of other features that are already available In Game.
In summation: I suggest trying out some of the existing adventure maps to see just what amazing things are already possible and being used on a regular basis. You might be surprised to see that our creativity is pretty well enabled.
I agree with everyone else, even if you want to do it on Vanilla Minecraft, you have to download the map to play it, and yes i know how little time it takes to do this, but you can't get all the commands and scripts you want.

Command blocks are amazingly useful if they are used right.

You obviously don't realize what the Command Block is capable of - go download and play Hypixel's map.
All that is done with command blocks. You'll be amazed.
Wool doesn't use greyscale, it has individual textures for each colour. I do think we need a bit more items for customization, like mob rails to specify a path for mobs and other things of that nature. But other than that, command blocks cover pretty much everything. Seriously, if you need a good example, look up Herobrine's Mansion by Hypixel, it uses command blocks BRILLIANTLY.
These ideas aren't too bad. But command blocks will do just fine if implemented right. Just get some mod that allows more commands for the command block, and it'll work wonders.
