A super-rare block that exists only once per generated chunk. The texture is generated randomly in the same way as the terrain, making each one unique. I don't completely understand how the chunk generation works, so the algorithm may not transfer--but the idea remains.
This would give players a more noble pursuit than the utilitarian harvesting of minerals. It also adds the the uniqueness and individuality of their world; one of the defining features of Minecraft itself in my opinion is the feeling of ownership one has upon generating and developing a world. It would be a difficult but rewarding task to collect the Metagem on each chunk, but players could take pride in their collection.
Here's a rough (very rough) mock-up of what they could look like:

(imagine a room full of completely different versions of these...)
Now there are many variations that can be done to the idea. The block could be semi-transparent showing a unique matrix of colored voxels. The metagem could be crafted into jewelry using gold and other metals/minerals. The block could even be indestructible, forcing the player to create elaborate tunnels to access or show off their discovery. The possibilities are definitely intriguing. I was thinking that the metagem could be nestled in a diamond cluster for even more greatness. Who knows?
Anyways, this is a fantastic game, and anything Notch can do to enhance that trademark Minecraft hey, this is my own world feeling would be well received. After all, isn't this why we play?
Yeah, there really should be some unique items that define a map, in another thread someone suggested making random statues and trinkets that can be very rarely found on a hell map. this could probably combine with that (metagems on earth, trinkets in hell).

I agree. I just think that this is an effective way of having truly unique items in each Minecraft world. I also think that having more things to do/look for while underground would be great.
