Hopper filter as mentioned, or item sorter. There are plenty of ways to make those and a simple youtube search should show plenty. Without mods, you generally can only sort items that can stack though, so keep that in mind.
When they walk into the room with the boss you can have command blocks go off with...
scoreboard objectives add Kills Killcount
Unless the boss has other mobs with it you can do
testfor @p[score_Kills_min=1]
on a clock with a comparator coming out to open the door. If you have multiple mobs in this room change the "min=1" to however many mobs are with the boss. I hope is helps!

Might be exactly what you're looking for. Have the boss drop a unique named stackable item which the filter waits for. Any other item could be passed through the system to some lava, but the boss item could set off the circuit that opens the door.

im gonna try that now
EDIT: It said "invalid objective criteria type 'killcount' ??
Edit: im not getting this whats this going to do once i make it work?
That vid shows more like an item sorter i Think thats not what im looking for im not sure
Well, I meant that as more of a starting point. I know you can rig it up (pretty easily, actually) to send out a pulse when the filtered item successfully passes through.

Just check out a youtube vid that'll teach you how to create a hopper. It's the way me and most people learned! If you still have questions shoot me a PM and I'll be glad to walk you through it

Really? Were you at the correct part of the command?
i wrote i na command block: scoreboard objectives kills kill then pressed tab and it did nothing
Oh sorry it onlybworks in chat to make sure you using the right objective might want to use the that.
