Compatators can output signals of varying strength depending on how many items are in the inventory it's facing.
I am guessing that's the problem here.

One item in a hopper does give out a signal. But it only goes one block. So you need a repeater after the comparator.
EDIT: Also items falling through hopper go much faster than items traveling in a hopper line. So it could be going too fast to send a pulse. If you need your stuff to go the chest at the bottom and that hopper to send out a signal, then I recommend putting another hopper next to it, which would then take it to the chest. So the item would slow down in the corner to be able to give out a signal. If you don't understand perhaps I can build a demo and take a SS for you later when I log into the game.
EDIT2: So this set-up gave a signal every time(tried like 10 times) No need for corners. So simply add repeaters after your comparators.
