I am designing an Iron farm for Minecraft 1.4 and I would like to know if the centre (16x6 Square) of a NPC Village is determined by the houses or by the villagers.
I understand that this is something not commonly known, so any help is appreciated!
I'm assuming, like Party said, it's centered around the well.
If you look at the layout of most of the villages, they're normally formed around that well which is more than likely, your center point.
I doubt it's the doors as you would have multiple center points and it wouldn't make much sense.
It is most certainly NOT determined by the well. You don't need a well to have a village. I would suspect that the center of the village is the center of an imaginary rectangle who is bounded by the valid houses. I imported villagers by Minecart into my town (which was not originally an NPC village) and the iron golems were originally spawning inside a building right in the center of town.
Mojang stated that the well is indeed the centre of the village. Then for the amount of villagers to spawn is determined by the doors in a 50x50 block radius from the well. And you do have to have a well in the village for it to be a naturally spawned village. If made custom, the game will take the radius of all the houses in that area and find the centre and mark it as an invisible well. Then do the same with the 50x50 block radius stuff.
In a generated village the well will be considered the center of the village. EVERY village has a well. Even the smallest. I once found a village with one small house and a well, that was it. In a man made village, the villagers will choose a random spot within the village.

The well might be the starting point for the village generation, as in the game generates the well first and then generates other structures based on the location of the well, but that does not make it the center of the village. A village's center point, for purposes of determining the village radius and iron golem spawning zone, is the geometric center point ("average position" or "average coordinates") of all the doors in the village houses.
As to whether it's determined by the doors or the villagers, it's the doors, but remember that a door only counts as a "house" if there's a villager nearby to "see" it. So while the villagers don't directly affect the center point, they can indirectly affect it by either recognizing a door or not, causing it to be or not to be counted in the average position to determine the center point.
