I have tried to make an aquarium in creative and followed the 20X20 block rule and used the eggs to spawn my squid. We'll it don't take long for them to despawn. Can squid not be kept in an enclosure?

No, squids are the exception and will despawn.

Just out of paranoia, try 19x19. There are times that MC's math seems a little odd, so that might be safer.
