Yeah, they really need to fortify the code for animal despawning. If an animal is stuck on one block, it shouldn't be protected from depawning. That's pretty frustrating, seeing all those cows and sheep mooing and bleating in difficult places to go kill them.
On survival island maps in any update, no animals spawn ever. They also don't spawn in deserts. So, my first question would be, in what biomes are we talking about here? As Cobra mentioned, if you have a large number of animals trapped in areas where they cannot move, they won't despawn and the animal spawning cap in the world could have been reached already. The same applies if you have a number of farmed animals inside pens. It is really more likely a matter of fully understanding the mechanics built into the game of animals spawning and despawning than likely to be a sudden "bug" in TU12. If you have an extreme hills area of the map, mine and move some blocks around so that you provide the trapped animals that spawn on cliffs different ways of walking out of their predicament. You could also go into the wild areas of your map periodically and clear out (kill) any trapped animals you find.

As stated above there are few animals in the extreme hills but the Forrest snow and swamp biomes are bare.
Edit: in the Forrest snow and swamp it is dead quiet when it comes to animal sounds and yes sound is working cause I hear them when in the small extreme hills.

Kill the animals stuck on the mountainsides and then rush back to another biome to see if you trigger some animals spawning there. The most likely scenario is that you have numerous animals in various trapped circumstances and you need to kill them to free up some spawning room in the animal world limit.
Yours are either stuck on extreme hills' ledges, in caves/ponds, or you live in an ocean/desert biome.
