Assuming that I build a new map and explore at the least a quarter of it, is it possible for a jungle biome to spawn in an undiscovered area of the map? Or would a new seed have to be generated when the update is released?
I haven't been around MC long enough to see a previous update be put in effect.
Yes, it's possible, although, there is no confirming a jungle biome will spawn in your seed. As well as that the new chunks will generate oddly and many of the world's biome will have relocated. You're better off starting of a new world.
Although, there is one thing I think might work. If you got a PC seed from 1.2 (since TU12 is essentially the PCs 1.2 version) and you use that seed before TU12 and it has a Jungle somewhere, then a Jungle should generate once TU12 comes out. Provided of course that you do not explore the place where the Jungle should theoretically spawn after TU12.

Just leaving a portion of the map undiscovered doesn't guarantee anything. The seed, as MustacheGuy pointed out, has to already include a Jungle biome within the limited xbox map. Currently, the Xbox lacks the code to generate Jungles so other biomes fill the void (to over simplify it).
In short, you're better off seed-searching for a jungle after the update come out.

The seed -1617224717 spawns you on sand a few blocks outside a massive Jungle Biome in 1.2 on the PC. So if one were to use the seed now and explore the opposite direction of the Jungle and then return once TU12 comes out, there should be a Jungle where they didn't explore next to the spawn.

Actually, possible means it can happen. Probability relates to the chances of it happening. Read a dictionary before you try to correct someone. It helps to know what you're talking about.

Right next to the spawn point? Then it almost certainly will not. If its within your view distance it is most definitely explored, even if it doesn't show up on a map. My rule of thumb is, anything within about a third of the map size in any direction from where you've been should be considered explored/generated.

Not to mention, if you're like me and have a tendency to run around w/o your map first, then double-back to fill the map in, you end up with a lot of explored (i.e. already generated) area that is not on the map.
Also, don't forget we don't have, and may never get, large biomes. In my experience, it's kinda uncommon to have a biome that's more than 100 blocks in any direction; which means, for most biomes on the 360 edition, once you're next to it, most, if not all, of that biome has been 'explored'.
