I'm gonna go ahead and say now, there's no 100% full proof way to defend a base against other players. Unless you have really trustworthy friends, and set some rules while playing, they're always going to be able to get in. The 3 main ways I could think of them getting in, other than the door, would be to dig under, build over, or dig straight through the wall.
To solve digging under, you could either build an 2-3 thing wall of obsidian going all the way down to bedrock. Which would both take a while, and take a lot of resources. Or, you could make an entire floor of lava under your base, so if they dig under, and dig up, they bump into the lava, and hopefully die. There are ways around lava though. And even obsidian can be broken.
As for building over.. There's not much of a way to get around that I don't think. Even if you built it all the way up to the high limit, they could simply build up the same high, and just jump over the wall. Lava would be another idea here too, but even then, they could just bring water, or blocks, and get around it.
Digging through, pretty much the same idea as before. Either a really thick wall that takes takes ages to break. Or a wall with lava inside layers of wall. But again, if you can build it, they can get around it.
The mechanics of Minecraft pretty much guarentee that they can get in no matter what. It's just a matter of how annoying you make it for them to do. Digging under and digging through the wall, you could make it rather annoying with a lot of lava and obsidian. Building over, I'm not sure how you could fix that, unless you make a 3-4 block thick obsidian roof on everything.
On the other hand, if you just wanna make sure they don't steal anything.. There's plenty of tricks you can do to hide a chest, either with redstone, or glitches. Just search youtube.
If you REALLY wanna be safe. Don't let them on your important world. Make a separate world for them to play on with you, and one just for yourself in single player. So you don't have to worry as much.
Those are short range for the most part,and waste a lot of tntAutomatic arrow dispenser,eggs,splash potions of harming,fire charges with infinite,and automatic firing.......anyone?
If you are trying to hide materials, private message me. I can make a vault that you open by walking into the wall, no touching anything, plates, switches, torches, buttons, tilling certain blocks, just walk into the wall. No one will see it.
If your hiding from MOBs while looking classy, make your castle have 5 thick walls. Outer layer, stone brick, or whatever you want, inside that, obsidian, then lava, then obsidian, then the inside wall whatever you want the appearance to be (stone brick for me.) so its outside on both sides stone brick, inside those on either side, obsidian, then in the middle, lava. That way the walls are wide enough to walk on comfortably.
For ultimate simple defense, just dig a trench 2 wide around your house 4 deep, MOBs don't jump. If that isn't enough, every other block place sand, and plant a cactus. 95% of the time a MOB won't get between the cactus's. If placed on the inside part of your trench, spiders climbing out are 1.5 blocks wide, they will either die or be extremely weak after climbing up a cactus.
If you want more ideas, just hit me up anyways. I do only survival mode on hard so we come up with some crazy stuff.

Well as the original post suggests protection from actual players rather than mobs, as has been said...if you can build it, someone can bypass it. Hiding stuff is the only way to protect your shiny stuff.
Chests under lava is one way. You can still open it with lava on top and access it. I used to do the trick with the painting over a gap in the wall, but too many people wise to that now, plus you can see through the gap at the edge.
Secret rooms at the bottom of wells. Set the depth so that you only just survive without drowning. But anything built will arouse suspicion and interest. So head into the woods, dig a hole and bury it. Keep a note of the coords. Someone might chance across it but the odds are against it.
what do you mean "defense"? like armor defense or protecting your house defense?
For Armor, (if you are going to add in new armor) you could rather extend the chestplates up or further down you health bar. or you could make a new texture for another chestplate that shows it's enchanted
For Defense like your house, you could make a redstone trap with buttons and you have to hit the right combo on buttons and if you get it wrong then you fall in lava! that way creepers or skeletons cant get in!

Unfortunately as I have discovered there is no good way to defend from griefers the best defense is to turn off trust players.
