There's no difference with or without Optifine other than both ways are wrong, but you should be using MCPatcher.
I am using MCpatcher, By without I meant with MCpatcher.
Anyway, This is a great texture and I hope youll get more items and block out!
Oh, if you do have MCPatcher and Optifine working together you should just get a FPS boost from optifine.
I'm hoping to finish /blocks/ somewhat soon because there isnt all that much left in there, then I'll be focusing on /items/ (and possibly start mobs too at that time).
Why can't I use optifine instead of mcpatcher? Doesn't optifine already include a type of mcpatcher?
EDIT: Forgot to mention that your texture pack is awesome! Its like a new and very much improved Sphax, I guess I should say goodbye to Sphax, since yours is way more superior!
Also your page is really confusing to me, what does PMC mean, which is the correct download link, could you make a bit more clear, thanks.
optifine doesn't has the same connected textures capabilities as mcpatcher, therefore you can't use it instead unless it's necessary (it's possible to install optifine with mcpatcher though)

How can you install optifine and macpatcher together, do you just patch one file and then just add optifine/forge/ is it that easy?
I only did this back in 1.5.x, I just selected optifine as a mod in the mcpatcher option to add and installed it, worked okay despite everyone saying that it wouldn't lol

I can't confirm it, but I have been told you can install MCPatcher and Optifine Light together, but I don't use Optifine at all so I have no idea how to go about it.
Love the pack and artwork, Also only pack I know of using the Connected textures mod to the max. Look forward to what your mobs will look like when you finish all the items!
