I want to make a 32x texture pack but I don't know how to get the 32x template.Please help.

Just resize the terrain.png in the minecraft.jar to 512*512 without any resampling methods turned on (if you do it right, it won't be blurry). Then paste on your textures as needed.

That is for 16x. And yes, just resize the image.
