A good start would be by editing a pack that is fairly easy to pull apart, such as Painterly's full-complement pack. That shows all the textures, skins, armor models, etc. so you can go completely overboard.
When you feel that you have a grip on editing 16x16 textures, you can go up from there. It is simply a matter of using the HD mod (stickied in this forum section) and bumping up your texure resolution to 32, 64, 128, or 256.
For editing, I recommend using Photoshop, as it makes editing and transparency a breeze, not to mention the filters and effects you can apply to your work. To be honest, I use Photoshop 6.0, but even back then it was still a powerhouse. (Basically, don't think you need CS5 to do anything good. Afterall, this is just texture editing.) :smile.gif:
