well i would recommend straight off that you use the free picture editing program paint.net. Its free to download and easy to use and also has transparency which is necessary for minecraft. As for what texture type to use for your wood you just have to try and make one that matches the rest of your pack but some simple things you can do if you are still going for a 16x version but not really anything too much of an overhall you can try just changing the hue/saturation and also the brightness/contrast. Those can make some interesting changes without you having to do anything to radical to the actually pixels. If you want to change the actual pixels to make a custom image you just want to make sure that your block doesnt seem like it has too many lines or spots going through it that make it not look uniform and nice. You also want to make sure that the block looks good when beside other blocks of the same type. If you arent careful they wont look that good when you increase the number of that block. Also if you can post a picture of what you have done on your texture pack i may be able to give some more advice about what you should do for the wood.
