download the texture pack of your liking then take the zip folder it downloads and place it in your texture pack folder

search for %appdata%
then open the .minecraft folder and the texture pack folder is there
Generally your texture pack folder will be somewhere like c:/users/*yourname*/appdata/roaming/.minecraft/texturepacks
The AppData folder is a hidden folder, so you'll need to show hidden folders to see it.
You can copy 16x16 texture packs to the texturepacks folder (don't unzip them first) and use them from the button in the Minecraft menu. If you want to use higher resolution packs, you'll generally need to use a patcher to modify the default texture pack.
Edit: The patcher is here ( viewtopic.php?t=46173 ) in case a link isn't included along with a pack you want to install. You should only need the patcher if you're using packs with higher resolutions (32x32, 64x64, etc)
For the sake of simplicity I will assume you have bought the game and have the latest version.
From the game menu select the Mod's and Texture packs option. This screen allows you to choose the default Notch created textures or any custom pack you have placed in the Textures folder.
At the bottom of the screen click the button to open your texture pack folder. Place any downloaded texture packs in this folder and they will appear in the list.
Note: Pay attention to the resolution of the textures you are downloading. Nearly all pack authors will clearly state the size in multiples of 16 (32, 64, etc..). If you use textures larger than 16px without using an application to patch your minecraft, you may see graphics glitches or even crash your client.
If you are just starting out and looking for your first pack I have a few to suggest. I will have to add more once I get home though:
Axepack - Hand painted, warm character with nice original style. Very well done and highly recommended. Shortstuff's Clean Design - A simple and clean redesign of the standard. Very well done.
