well for a 16x16 get the original pack out of your .jar and edit it as you please then save and put it in a .zip which you then place in the texture packs folder... be sure to use a program that supports transparency, I advise paint.net unless you are rich then photoshop CS5, paint.net is free and supports transparency the paint your computer comes with does not.
I always tell people to do what I did. Play around a ton. Whatever you edit in the terrain.png, test and see it in game. Make silly faces on the blocks or past your own pic. Then see it for yourself and you'll so what to do or not do. Keep in mind the size of the blocks and don't stray out of the lines. Use a terrain file that you like as a judge for each block.
But yeah, Photoshop if you got it, Paint.net if ya don't. (I use Fireworks too)

Here's both original terrain pings. One is 16x16 and the other is 32 by 32. I would suggest the 32x as there are SO MANY 16x16 that appear and disappear. Plus you get a better chance to make it stand out.
