If you're serious about making a texture pack I suggest you first get familiar with, well, art in general. You won't be able to use MS Paint for the retexturing due to it no supporting transparency, so either get Paint.net or GIMP if you're looking for free alternatives. Get familiar with how to use them and then use them to re texture the packs by simply dragging the files from the .jar file into them. I strongly recommend you actually practice some basics in those programs before you start anything like re texturing a game.
Alright, first, get Paint.net, it is awesome for pixel art. Its also easy to learn and easy to use. Now, if you dont have a program named Winrar, download it and you can continue. Open the Minecraft.jar with Winrar. I would suggest you also make a texture pack .zip in the texturepacks folder of the .minecraft accesed by running %appdata%. Now extract the files you want to edit from the Minecraft.jar into that new texture pack .zip. Edit these textures using Paint.net and save it. In order to actually save the files in the .zip, go to the zip of the texture pack, and say yes to if you want to save the new thing into the .zip. Now run Minecraft. Choose it from the texture pack list and run a world.

Could I also use a preexisting texture pack and edit that instead?
