Some people play by using inventory editors, some people play on hard, others peaceful, and some like to fight. So post the way YOU play.
I like to play on hard mode, never using any third party program (including texture packs and mods), and go for my resources at night hoping I don't get blown up or shot at, while living in my house adding to it in the day.
I just try to get as much wood as possible in the start, make a base and hide in it at night, come out in the daytime and collect more wood. I try not to use any 3rd party programs at the start but then I get super bored, use MineEdit and make castles out of Diamond blocks!
I play normal, no mods, no texture packs, no third-party programs. The only file manipulation I do is regular backups for when Minecraft forgets how to work properly and corrupts my saves as it has done multiple times.
No mods, invedit only for my sandbox world 5 or crash recovery. No textures. Peaceful.
Favorite thing to do when bored? Stack of each: Sand, Sticks, Logs, ladders. 4 Stacks of torches, 5-6 iron picks a diamond shovel and a bucket of water. Lock the door and go spelunking for 5 or 6 hours, coming back loaded with goodies.
Its amazing how long one stack of logs will keep you sustained underground. Sand makes a great rappelling rope if you find a very long shaft, as well as a marker.
I play with two hands most of the time. If I'm just traveling, I use only one.
I start by using one hand and clicking the button I have to open the game on my quick launch bar, then with my other hand I hit space to get past the java running confirmation. From there I mash my keyboard to enter a password (it doesn't matter since my laptop isn't connected for the most of the time currently anyways). Then after accepting to play offline, I choose single player and whichever world I wish to have my presence in. I then build as I see fit with, what I can acquire through in game means.
I play with no texture packs(I think the vanilla textures are better than every other pack), NO MODS EVER AGAIN(I tried installing one, ONCE, and I ended up temporarily breaking minecraft), I used to play on peaceful because I almost was stuck outside at night on my first night, so i freaked out and started playing peaceful, and got stuck on it. I started playing normal recently on most of my worlds(explained later). I start out by gathering wood and making a simple wood house. After I get enough stone I build a cobble house. Then I create a mineshaft and branch mine.
There is one world I play on, 5, where I use INVedit, it is a place where I test concepts, test redstone, create things almost impossible in regular game(Such as giant statue of a creeper(Thanks Lukedude5) made entirely of mossy cobble), and just have fun. I never used INVedit for any other save besides this.

To be honest I play on Peaceful, I mine and mine and mine and amass massive amounts of coal, Iron, Cobble and Redstone along with a respectable amount of diamonds, all in preparation of some vast project which is not yet even forming in the depths of my mind, but one day i'll either use all those resources or just trash the save and start from the beginning all over again.
I play on normal(sometimes hard if I'm bored), almost always vanilla with occasional use of ambient occlusion. Most of the time I just wander around on the surface looking for mountains and resources (wood, coal, and the such) since my maps recently have all been huge flat desert biomes. Use of inventory editors and mods restricted to my World 5 where I mostly end up destroying a part of the map, deleting it, then starting it fresh for other testing.
I play normal survivor. Clear cut a forest on my first day to get a few stacks of logs, then hollow out a small room in the side of a mountain/cliff. Make tools/doors/chests. By the time that is done its usually night time and I begin my tunneling deeper into the mountain. Currently don't use any texture packs/mods. Don't plan on using any edits/hacks unless I'm just messing around and testing things.
I'm very new to minecraft but right now it's mostly about extending the land around me that's safe from attack.
I play on normal difficulty. No mods. I made a base on the border of forest and desert. Found a cavern below me that I explored and mined before closing off. I've been having fun tracing another cavern under the base's inner courtyard. I've been revealing it up top sealing it off with glass so you can look down into it. There's a glass building that sits on an entrance I made to the cavern. I've been working on enlarging the buildings of the base. I also recently made a waterfall in the courtyard to try to beautify a bit. I've got a mine nearby. Also I've got rows ready for wheat if I need any more and I'm working on making a tree farm area for more lumber. Eventually, I'd like to make a portal.
I find I like both single player and multiplayer for different reasons. In multiplayer, it's very fun working on stuff with my husband but I really like the feel of accomplishment that I get from single player.
