Well, Ghasts can destroy blocks, so a closed room isn't really useful.
And, to answer the origional idea, portals must be set on fire, so beat out the fire, and the portal goes bye-bye
The problem with that is that once set on fire, the flaming part turns into the purple portal part. I doubt we can extinguish the flames if they are no longer there, but it'd be nice if we could.
