I like this general idea, that in the future some very "special" weapons be made that can be used to defend against an otherwise nearly impossible enemy.
However, in least in my experience, redstone is a wee bit common to be very special. Not that its particularly easy to find, but after about a week of hardcore mining I have multiple 64 stacks of the stuff. Compare that to maybe finding about 1 or 2 dozen diamonds total since playing the game for the first time.
Take what you want of my opinion, just throwing it out there.

Yeah, um, no. I don't think Ghasts should have a glaring weakness. I want them to be something that you're going to run away from unless you know what you're doing, not something that you just grab your weapon (regardless of rarity, you can always hold off on building a portal until you have the item to deal with ghasts; you can never be absolutely sure you have the skill to handle them until you try, if there is no secret weakness) and charge at with a battle cry of "THIS WEAPON IS USELESS FOR ANYTHING OTHER THAN HITTING YOU!".
And I think it'd just be an irritating obstacle and an excuse to put redstone (or slime, or seemingly unused material #47) in crafting recipes that it really doesn't need to be in if the ghast was difficult with its weakness and impossible without it.
I do hold some intrigue for the gold sword idea, but I really think that gold's usefulness can wait until (if?) enchanting is implemented and we can add all kinds of effects to gold equipment. Then we could have an "undead killer" or "hellspawn slayer" enchantment to deal with ghasts, but I still think it should only be enough to be worthwhile, not enough to make it actually feasible to battle every ghast you see.
Hmm, I think if there is a way to deal with ghasts, it should be a purely defensive thing. Like a sheild that blocks it's fireballs, it could also block skeleton's arrows.
Though I'm thinking that the wood one would only be able to block one fireball before it's used up. Ghasts spit fireballs, right?

But yeah, I think shields would be really handy. If you could wear them like armor to completely deflect projectiles and take a sizable chunk off (or have a chance to completely block?) the damage of an attack, that'd be pretty neat.
Yes, ghasts do spit fire at you. Some mobs just have no sense of courtesy.
Considering that it takes obsidian to get to hell, why not make obsidian weapons that are like stone weapons to all regular enemies, but are like iron or diamond to the denizens of hell? You could even make ghasts invincible to everything except obsidian, just to make sure you can't use your diamond sword.
How about their weakness is water, since they are fire/lava based mobs? Then all we would need would be a water arrow.
How the hell would you shoot an arrow made of water? i know theres going to be alchemy but still. I think snow balls is there weakness. -throws snowball at Ghast- "aha!" Its super effective, Ghast faints!
I would like if you would need to make maybe a gold or obsidian sword to kill the ghasts, but then you would have to enchant it using a HUGE and very expensive rune, like maybe using 30-something iron blocks, like 5 gold blocks, and one diamond block. dimensions of maybe 30x30. then you could have something like:
Yes! now after days of hard work i can finally create my ghast-slayer!!!
It would be like creating this huge construction so that you could be relatively safe in hell and it would be a goal the you could constantly be working towards. I just really like that idea. oh and there would be alot of complicated redstone wires and circuits involved.
