Have 2 stacks in storage, now use all i can find quite liberally, full enchanted diamond armor, some picks for keeping etc. Useless waste of non-renewable materials, but meh...
Method? Creating underground railway tracks in diamond level, keeping fortune 3 pick ready. Just hack 1x2 tunnel to that mesa or desert or TaigaM or whereever 2000 blocks away, do it on level 12 and you soon find that iron is more precious than diamonds.
In my original world, I'm not sure exactly, but I ended up making a full stack of diamond blocks to save space. Currently I'm in creative where I've been going a little crazy with diamond blocks so it doesn't count. BTW, I have a reputation as the guy who "eats lava and poops diamonds" due to my ability to never, ever fall in lava, and for my ability to find diamonds accidentally.
In my single player world I have played it for about 1 year and a half and have 3 64 stacks and a 34 stack of diamonds UNUSED. Everything total about 4 stacks.

On my mcmmo SMP server, I'm sitting on ~5 stacks currently. I usually go down in the branch mine once a day and go until I find a node or two. Having 500+ mining skill in mcmmo + using mcmmo's super breaker ability + a fortune 3 pickaxe means a vein of 6 diamond ore yields anywhere from 18-25 diamonds
Strip mining is kind of a meditation exercise in combination with listening to music. I can easily get a couple stacks in a couple hours and a couple level 30 enchants from mining xp and the occasional cave exploration.

Wait, are you guys counting diamonds or blocks? If its diamonds, I currently have 337, but I'm pretty sure I've used about a stack on gear.
Funny thing is I currently have absolutely no iron. I just exhausted my entire supply making pistons for my endermen farm.
