I'm not a builder, so I don't have any suggestions for that.
However, for a single-biome challenge, I like Frozen River and Mesa best.
For Deserts, there's no reliable way to get wood above ground, which is required for survival, so I'm not a real big fan of those. It became a lot more viable in 1.9 when dead bushes started dropping sticks, but reliance on villages (or the RNG for a world seed) is the only dependable way to survive, and I'm personally not a big fan of that style. Still, I have a Desert-only hardcore game going.
Frozen River is a flat, infinite plain of ice with tiny little islands dotted here and there. Rarely, there will be a tree growing out of one, so gathering wood is possible.
To me, a Mesa single-biome world seems even hotter than a Desert one due to the Mesa's reds and oranges. It also generates a lot of Abandoned Mineshafts at the surface, so wood can be obtained that way.

What I think could be cool would be an Infinite Icespikes biome. Now from a survival standpoint, this wouldn't be a very good challenge because 1: it's so cold and 2: there really is no way to get wood at all. For a survival challenge, maybe Dark-Oak forest. Tons of wood. Almost always dark because of the trees, and so many woodland mansions.

I've been able to get wood from mineshafts, I wish I could find a sapling. Ive been to the nether, this is hard mode too. The real trouble is making a xp grinder.
Frozen Ocean. You pick a direction and start walking. If you don't run, you won't starve. You will eventually find a small outcropping and a tree, and you're off to the races. This first part is actually more boring than hard since mobs won't spawn on the ice. It's not my favorite by any means, but it really clarifies what can and can't be done.

Your post inspired me to give Deserts a try again. Single biome and Hardcore are the only two options I selected.

That's a really cool desert you got there! Would make a great spot for a nether portal or a mine entrance I think.
Honestly people, what is the best/worst experience ya'll have had on a one biome world, as in not normal. 10 creepers, lava from a roof, etc.
Mushroom biome!
No trees there. Or villages.
I gave up after spending quite a while trying to turn a dungeon spawner into a mob farm with no tools.
I had the same experience on a desert world where I didn't allow myself to use the trees I found.
I suppose I should have kept exploring to find more village chests.
I've done several desert worlds and never had any trouble finding a tree, eventually.
Just follow the rivers, when one runs out follow the ridges until you find another.
It sometimes takes a few MC days, then it gets boring hiding out in a hole until dawn.
