Say that I wanted to run on a superflat world. I want to go the furthest I can, and have 1 potion of swiftness with 1 piece of redstone and 1 glowstone dust. What should I brew? Potency or Duration?
Assuming you stop running when the potion wears off? I'm going with duration. One level of potency won't offset the extra time. The potency will make you even faster, but compared to the time increase... You'll get farther with duration I'm pretty sure.

Alright I tested this out in a superflat map. When I used both potions, I walked in a straight line, and marked each point in Rei's minimap. I also did not sprint either one. This might not be 100% accurate, but it does prove that duration is better.
With a speed 1 potion, effects lasting for 3 minutes, I was able to travel about 805 blocks
With a speed 2 potion, with effects lasting for 1 minute 30 seconds, I traveled roughly 450 blocks.
I was able to travel almost 2 times as far than with a potency potion.

But then how do you know that Rei's Minimap was giving you the coordinates you set!?
I marked each spot with a waypoint. When you open the waypoints list, if you hover your cursor over each waypoint, it'll give the x, y, and z coordinate of that waypoint. So I set one for the starting point, then marked the end of each run. After that, I'd add the coordinates that had changed (in this case, I was moving along the Z axis).
Starting point: Z:-432
End of potency potion: Z:19
End of duration potion: Z:373
After the end of each potion, I would go back to the start point, and walk forward again.
To figure up the totals, add 432 to the ending point (since you need to travel 432 blocks to get out of the negatives in this case). So:
