I wanted to start a youtube series where I start at beta 1.0 and continue on in the same world, but the end isn't added until the official release 1.0. If I start in beta and update to version 1.0, will any of the new chunks generate with a stronghold, or will I be unable to access the end in that world?
Yes, as long as you don't explore the chunks where the strongholds would be (between 640-1152 blocks from 0,0); you'll be safe if you only explore that far in one direction as there are 3 spaced 120 degrees apart, but you may have to go in the opposite direction to get eyes of ender to locate the others. Also, since 1.9 there are 125 additional strongholds (128 total) across an area up to tens of thousands of blocks from 0,0, so it is guaranteed that you'll never generate all the chunks they would be in.
Also, the only known case where a world won't get new features is when updating from 1.1 to 1.2, which will convert the world type to "default_1_1", which retains the biome generation of 1.1, which means there will be no jungles (this also breaks 1.7+ worlds, which will have no savannas and far fewer deserts due to Mojang not knowing what they were doing when they updated the biome code, with the biome list replacing the one for hot climate zones). You can however use NBTExplorer to fix this.

No. Like TheMasterCaver said, as long as you don't explore the area the strongholds will spawn at when you upgrade, you will be good to go. You can check this by simply getting your seed and going to one of the many websites to find the exact place (x,y,z) your 3 strongholds (from 1.0-1.8) will spawn at. As long as you stay away from at least one of those areas to avoid loading in the chunks, you will be fine.
