I have always wondered. Since the default minecraft textures are 16x16, will using a 16x16 texture pack cause fps drop?
Don't know if you meant this or not, but you're comparing 16x to 16x; no difference at all.
Anyways, when you increase the size of a texture, the file gets larger and thus the game has to store more data in memory. With one or two blocks, this isn't bad, but with an entire texture pack, depending on the size, it can decrease FPS dramatically. The amount of lag that occurs is highly dependent on your GPU, with a low-end GPU, a 64x pack may be too large and may lag down the game pretty badly. However a high-end GPU can go to 128x and higher with a smooth 60 FPS.
So, the answer is; it depends on your computer. Either way, it will drop frames, but the exact magnitude of the lag depends on your computer.

I don't see why it would... If it does, it wouldn't be noticable
Although using special textures, such as connected textures, or random mobs, you might feel a slight decrease, but very minimal.
