I actually just went into the Nether and came out with about 70 piles of glowstone dust (and like 30 red shrooms. Man I'm happy). I just dug up until I found a large platform near the ceiling. So just stay on land and go up and you should be good.
Well, once the Ghasts spot you, you're pretty screwed, so I think stealth is the key. Haven't tried any of the following ideas myself yet, but they're what I intend to do:
-Have a general hiding spot. Ghasts will only follow/shoot at you if they have a clear line of sight, so a building made of resistant materials should make you invisible to the Ghast. Also, the Ghast's fireballs are actually small explosions, and are the weakest "explosion" type in the game. Basically, this means that even Cobblestone can withstand a Ghast fireball (though a wooden door can't, so make sure you have an iron door). Check this page for a full list of blast resistances (the fireball is about a 20)
-Stay out of sight while mining. A Ghast's fireball may be able to destroy Netherrack and Dirt, but they can't see through them. In fact, Ghasts can't see through Glass or Portals either. The only block (other than Air) that Ghasts can see through is Lava. You don't need to completely seal in the Glowstone, you just need to block the line-of-sight from further areas.
-If a Ghast spots you while mining, RUN! The fireballs can still destroy the Glowstone deposits, and keeping the Glowstone intact (and holding onto the Glowstone you've collected) is more important than defeating them.
-As for how to stay alive while running away, here is a direct quote from the minecraft wiki article on Ghasts:
Ghasts aim at the camera, not the player. Going into 3rd person mode and having a ghast fire a fireball at you will go over the player, given the right camera angle.
The main reason I haven't tried these myself is that the location of the Portals I've made so far made them unnecessary. The first Portal I made was on a tiny island of Netherrack above some Lava, which didn't last long around the fireballs, while the second Portal took me to the roof of the Nether, in a cave right next to some Glowstone which was already mostly sealed off from line-of-sight. I'd appreciate it if you guys could give me some feedback on how well these strategies work.

The best bet is to just explore around and find clusters of glowstone that are close to the ground. If you find caverns or enclosed areas, those are where you should look first so you can get away from Ghasts.
It's not hard to find glowstone, because it's abundant in the Nether. I never mine it if it's in an open area and more then 10m from the ground.
just make sure that when you are exploring the nether, bring torches and wool carpet to mark your path. always some extra obsidian, flint and steel, compass, and bed in case you get lost. Worst case scenario, you can pop out somewhere in the Overworld and trudge your way back to world-spawn.
if you found a village already, priests have a chance to trade glowstone.
I'm not sure why the 3 year old thread was bumped.... But since its here, did the nether change since 3 years ago that much? I frequently see glowstone in semi-sheltered low-ceiling areas and just get it there when I'm exploring in the Nether. I see no reason to climb high pillars to get the high stuff.
